9 .   Optimization of HS/GC-MS Method for the Determination of Volatile Compounds from Indigenous Rosemary

Abstract:

The volatile compounds from rosemary dried leaves were extracted and than separated and identified by gas-chromatography coupled with mass-spectrometry. Different quantities (0.15g; 0.5g; 1g) of dried rosemary leaves were incubated at various temperatures (30oC; 40oC; 60oC; 75oC; 80oC; 95oC) and periods of time (15; 30; 40). A number of 4 to 28 compounds were separated and identified from rosemary dried leaves, depending on the used method. In all cases, the major compound of rosemary was α-pinene (46.52 72.45%), followed by camphene (6.54 - 14.03%), 3-octanone (2.97 7.84%), and eucalyptol (2.94 8.87%). By increasing the incubation temperature, the incubation time or the quantity of necessary sample may be reduced, the obtained results being similar with those obtained for the same incubation temperature but for a larger incubation time or quantity of sample.
